Welcome aboard, Wayne ! That's quite a few questions, all of which have answers in very detailed threads here. I've grown Blue Java bananas (etc) for 3 years now (with over-wintering) fabulously. Removing & re-planting pups is fairly easy. Simply remove a small piece of the corm with some roots to keep the pup alive. This does Not hurt the mother plant as long as you cover the cut with dirt. BTW, I learned this from a Hawaiian Agricultural DVD (University of Hawaii) that you can find here in the forum.
Good things to check out are the difference between Sword & Water pups and the pup separation threads which go into detail with photos.
As for Tissue Culturing, I know next to nothing BUT we have an entire section (& experts) devoted just to that!
