Shoot-tip culture
I have received a few emails/IMs requesting info about shoot tip culture of banana. I have re-done some of my original Banana Micro-propagation page. Feel free to visit it. It is still rough and only about 1/2 of the info is there. But there are some pics that answer the most common question I get, which is: what does the explant look like and how do I cut it out?

I'm trying to make a flow cabinet out of an old fridge. I'm not sure if I'm going to use vertical or horizontal flow though. My fear is that horizontal, blowing towards the front may cause uneven air flow because the filter is much smaller than the work area. Vertical flow just means more work to install but may give a more even pressure. Any ideas? Anyone? Anyone? Bueller? Bueller...? |

Excellent page, very informative!
But I have one question: After you use Phtotechs Multiplication medium for initiating the culture, wich one do you use for rooting? I don't think Phytotech sells a medium without the cytokinin? Phytotechlab really confuse me: Does the banana-multiplication medium really contain EVERYTHING (just add water as you say), or do you need to add more hormones etc.? And what then, is that other product called "banana basal-medium" for? Erlend |

I failed miserably with my TC-trials this winter, but I suspect the explants were too small, and too weak. (Basically seedlings that have been sitting in the dark). I will have plenty of strong suckers in August-september to try. Would you say some species are easier to TC than others? Erlend |

If you hit reply to this thread, or when creating a new thread, etc..
There's a button that says "Manage Attachments". Click that and then you can proceed to upload the ppt directly to your post for people to view on their computer. Cheers |

Thanx for your help Keith! My filter is 2'x2' so I guess that's what I'm building. Do you have any suggestions on the depth?

Keith, how often do you typically subculture a batch of banana-shoots? My aim would be to maintain a collection throughout the winter.
