Shoot-tip culture
 
I have received a few emails/IMs requesting info about shoot tip culture of banana. I have re-done some of my original Banana Micro-propagation page. Feel free to visit it. It is still rough and only about 1/2 of the info is there. But there are some pics that answer the most common question I get, which is: what does the explant look like and how do I cut it out?

Great info, thanx!

I'm trying to make a flow cabinet out of an old fridge. I'm not sure if I'm going to use vertical or horizontal flow though. My fear is that horizontal, blowing towards the front may cause uneven air flow because the filter is much smaller than the work area. Vertical flow just means more work to install but may give a more even pressure. Any ideas? Anyone? Anyone? Bueller? Bueller...?

Excellent page, very informative!

But I have one question: After you use Phtotechs Multiplication medium for initiating the culture, wich one do you use for rooting? I don't think Phytotech sells a medium without the cytokinin?

Phytotechlab really confuse me: Does the banana-multiplication medium really contain EVERYTHING (just add water as you say), or do you need to add more hormones etc.? And what then, is that other product called "banana basal-medium" for?

I would go with w/either m519 (adding 2 gm/L gellan gum and 30 gm/L sucrose)or L467

Yes. Just use as is. If you want to add more BAP, say up to 10 mg/L you can, but I wouldn't bother.

It is just another formulah, but from what I read, not as popular as the one from Vuylsteke.

Excellent info Keith! Can't wait for your page to be finished.

I failed miserably with my TC-trials this winter, but I suspect the explants were too small, and too weak. (Basically seedlings that have been sitting in the dark). I will have plenty of strong suckers in August-september to try.

Would you say some species are easier to TC than others?

Use larger source materials. The greater the distance from the outside to the meristem, the less the contamination - as a rule. And harvest the suckers shortly before you use them, sitting around invited contamination.

Yes, and different groups behave differently. Plantains tend to form cormlets, where dessert bananas tend to form shoots. Some are very slow, others proliferate with abandon.

This is prolly way to large for what you need. Look into a smaller plastic hood and simplify the design.

A valid concern - but if the filter is sized to the chamber, it is a superior design.

Resize the work area and go horizontal. JMHO

Thanks - there are several areas I really need to flesh out, particularly sterile technique. I think that trips up people in the beginning.

I also have a power point presentation I was supposed to give at the CRFG fruit festival several years ago. I couldn't make it as my mother was having some health issues. Ian McGuire gave the talk, but I still have the ppt if there is any way to post that on the web. (I am sure there is - but I haven't a clue)

If you hit reply to this thread, or when creating a new thread, etc..
There's a button that says "Manage Attachments".
Click that and then you can proceed to upload the ppt directly to your post for people to view on their computer.

Keith, how often do you typically subculture a batch of banana-shoots? My aim would be to maintain a collection throughout the winter.

Depending on the cultivar and the temps about ever 4-10 weeks, with every 6-8 being most common.

20 inches seems popular. I would think 18-24 would be good.
