Working with 10 liter MS packets
Hi, everybody. I should FINALLY be receiving my supplies this coming week.
Would I have to prepare the ten liters as soon as I open one packet or can I store the rest in a ziplock bag or other? According to " Plant propagation by tissue culture " ( Thanks, Remko ), " Autoclaving changes the pH of media.. without sugars the change is small Most agars cause the pH of media to increase immmediately after dissolving... The pH of stored media tend to fall if they are stored... To minimize a change... should be stored in dark at 4ºC... So, could I just prepare the mix of MS ( 2 liters X 5 strength ), without sugar nor PGRs, autoclave and store in the fridge until use? Suggestions appreciated. |

I have stored it in solution in a refrigerator before, but still always had to adjust the pH after adding in the sugar, gelling agent and PGRs. When I did it at home, I bought 1 liter packets and just made 1 liter at a time without adjusting pH, but I also used very purified, non-mineral containing, water (Penta brand) so as to minimize any changes to the medium.

Ms instant media from such company usually do not containing hormone like BAP, so dont forget add it. in my lab i usually mix self all nutrition including makro, mikro, vitamin, hormon etc. |
