Trying to germinate usa veluntina in TC
Hello everyone i am trying to germinate Musa Veluntina in tissue culture and would like to know if anyone has a metho they could share.
Currently i have my seeds in a 1/2 strength Murashige and Skoog Media solidified with gellan gum. There is 3PPM gibberellic acid in the media as well as .1% Plant preservative mixture. and 1% sucrose If you see any problems with my current set up could you let me know |

that is not true at all at least not when Musa Veluntina is the banana in question
This study claims other wise and if you would like me to dig them up again i read three other studies claiming gibberellic acid helped germination of musa veluntina as well... I have not found a single study suggesting gibberellic acid inhibits germination Of Musa Veluntina and if you please provide me a link to one that would be useful as i would like to see what the conditions were besides the application of Ga3 |

For anyone wanting to germinate "musa velutina," it is simple as using jiffy trays, seed starting soil, and a Ziploc bag. It takes approximately two weeks but this was the method that I used. I had a 63.6% (7 out of 11) germination rate. I do not know if this would work for other varieties of banana plants to start seeds as I have heard they have been more difficult to germinate.

Klaus did you put the seeds in the medium as a whole of did you get the embryo's out (sterile)?

Hey thanks everyone sorry i got so snappy there just got offended a little to easily... anyways the seeds were put into the microplates whole after being soaked in warm water for 3 days. I'm starting to gather that it is mandatory to scarify the seed... does anyone have a good method for doing so? Also how would you go about extracting the embryo without damaging it??
P.s. the seeds were sterilized for 4 hours in a 5% Plant Preservative Mixture solution. prior to being placed in the medium |

here is a doc about GERMINATION OF Musa balbisiana SEEDS AND EMBRYOS (both english & french) i think that's exactly what you are doing with velutinas. but they don't explain how they cut the seeds longitudinally. (nail clippers ?) |

When trying thsi technique I used a pen knife, you need something sturdy to get through the seed coat. I've also read the embryo being squeezed out like a spot with a pair of pliers, that was with heliconia though.
