hi I am thinking about trying tc again third time is the charm or so I have heard
last two attempts grew callus tissue and turned green but went no further I used baby food jars and their original lids not vented is it necessary to have vented lids for gas exchange?
their is a possibility I did not get the proper meristem tissue in past attempts

No, you don't need to vent them.
Was the callus brown after all?
Maybe your PGR was not well dosed.

That's a nice piece of equipment. Looks like you are planning big jobs.
You callus didn't grow most likely because of lack of auxins. By the way auxins disintegrate when exposed to light and heat. So you need to change the medium more often.
Did you control the light and temperature?
With the right variables in place you should obtain 3-6 bananans in 3 weeks from the right callus. Then subcultured.

thanks again for your help
