Hi
l am going bananas, l have been trying to culture Williams and dwarfs and all l get is a single shoot. How do l make my initiates multiply-media suggestions that work?

What is your medium formulation, especially hormones?
Normally for shoot elongation very little or no BA is present, but for multiplication you need to have BA present in high quantity. In the lab I used to work at, for shoot proliferation we would use 1ml/L of 4mg/ml BA solution in the medium. You can multiply on this medium for up to 5 subculture cycles, at which point everything should be transferred to a medium with no BA and should elongate and root. After 5 cycles, you increase the risk perpetuating undesirable off-type somatic mutations, so you should restart with healthy, normal field grown stock.

While we are on the subject, can anyone recommend a decent kit etc. to learn how to TC? What is the best way to learn?
