I think its amazing see alot of baby pineapple in one bottle. i have alot bottle pineapple and this is for sample. is it awesome right?

Hi novisyatria,
I have a Pineapple crown going in a pot. Can I use that for an explant?
Would I use sections of the of the base of the crown, kind of like you would for a banana? I would really like to try this.

yes you can use the crown as explant. its very good as explant source. take the shoot (part tip crown). for sterization using bleach 10 % ( 10 ml bleach + 90 ml sterile water) for 10 minute, rinse 5 x with sterile water, and then soak in alkohol 70 % for 3 minute. rise 5 5x with water. and explant ready to planted. try media MS + 2 mg BAP + 5 mg NAA for shoot multification.

I am amazed at your caladium too. I have one of those growing too. I may just give that a try also.

i use agar-agar that we usually use for puding. alot in supermarket and cheap heheh. 1 sachet containing netto 7 gr. its enough for 1 liter media.

Hi novisyatria,
I didn't get around to trying the pineapple yet. I've been busy with other plants, mainly bananas.
I did a test with Gelcarin GP812 on bananas to see if it would help with the problem of phenolic extrudes. So far it seems to being working quite well. I was having problems with about 1 out of 3 cultures. Now, none are showing the problem.
Gelcarin is hard to get. It's actually cheaper than agar, $38 a kilo, if bought in bulk but, everyone jacks the price up to make it more expensive.
One nice thing about Gelcarin is that you don't have to heat it to make a gel. That saves some time. You do have to autoclave it to sterilize it though but no double heating. You can just add 7 grams in water; the stuff dissolves and turns crystal clear. Lovely stuff.
I do want to try pineapple, maybe this weekend. When I do, I'll post some picts of the explant I use. I'm not sure, since I have the crown growing in a pot of soil, whether contamination might have gotten inside of the plant. If so, I'll go out and buy another pineapple from the store and use that.
After I post the pict maybe I could get your opinion on how my explant looks.

Novi, just a question. Seeing how much you work in TC area, what are your average monthly costs in $. For everything... media, pots, lab, initial investitions... I kinda wonder, how expensive it really is in your area. Thanks.

you add food coloring/ oh no. food colors is poison for explant. your explant will die.

No, food coloring isn't poisonous to the plants. I have about 350 Musa Basjoo explants in jars and another 100 jars filled with different plants that all have food coloring in them. They are doing fine.

are you sure?
i ever try add food colors and the results not good. even there is explant can growth but they multification and proliferation limited. that why all scientist that work with tc never used food colors, except for souvenir for example.

Hi Novi! They look great!
Nothing is happening with my pineapple yet. I bought a pineapple this weekend and will give this one a try. I'm going to take a video of the process. I think I cut too small of pieces with the last pineapple. I did try to leave some hard base and soft upper section on the explants but maybe cut them too thin. Maybe I should make my cuttings a thicker and a little larger?
How long does it take for them to respond? Do you lay them on their sides on top of the media or poke them in?
Oh, I tried a caladium and a small shoot is just starting to show. :woohoonaner:

yes i think you should more thicker. for bananas respod to hormone about 3-4 weeks. but after have callus they wll growth faster and can sulcultured soon. congrats for your caladium explant. in what kind media? i did it with 1 mg TDZ. did you used coloring foods for this caladium? maybe better without food colors. i will post my new caldium callus in other thread

Hi Novi,
I'm good with bananas. I have a bunch of them going.
The pineapple, not yet. I put some of the slices vertically in the media and some I just laid on top. The ones I laid on top seem to be doing better. The ones I put in vertically, the tops have turned dark. Is that normal?

i put them vertically and they all good. i think its efefct food colors. for tc pineapples through callus induction more sensitif than shoot induction directly like bananas. so try with no food color.
part top become dark i think maybe they got browning. maybe you cut too small and tehre some bleach unclean or sterilieze too hard or too long. but my pineapple explant have not turn dark like your.

Hi Novi, My first attempt the explant turned brown and don’t look like they are going to do anything. I tried another one this weekend and they look better but I have wait and see.
When you cut the pineapple, and you see the dark circles where roots develop, do you trim that down further? I mean how far from the side do trim it? I’m afraid of trimming the side too much and removing the area where the roots form?
Oh, I didn't use food coloring on these:-)

do you mean cost for producing nanas/pineapples by tc? well more plantlet you get more cheap cost per plantlet. plantlet/seedling nanas its price about ` 1 US$/plant
